As someone who repairs computers for a living, I'm here to tewll you that turning them off and on actually stresses them. i "never" turn ine off u nless the power goes off or a program requires a reboot.

I left my laptop on for about 2 years straight and I feel like it did have some effect on it in some way. Fans running for 2 years straight, etc

I wouldn't be so sure; one could speculate in the exact opposite as well; that temperature-cycling the oils and bearings in the fans wears them out, or similarly that charge-cycling power-supply capacitors could have adverse effects, and such things. It's not as if I haven't noticed my constantly-turned-on computers actually seeming to have better lifetimes than those of various friends, who seem to have computers break for them every few years or so while I've still hardly suffered a single hardware failure.

Anyone can speculate, but painhertz speaking with such confidence made me wonder if he had something to back up his claim.

It depends. If you leave your computer on it will accumulate more dust and debris within the case and on critical hardware such as the motherboard. You really want to shut everything down once in awhile and clean it out to prevent this from negatively affecting things.

In addition to this if your computer has a sleep mode this eats up system memory and your computer will allocate HDD space to handle these processes. Disable sleep modes to prevent this from happening.

Otherwise, though I've never heard any scientific backing on this, supposedly leaving the excess bit of electricity in your system to be idle can damage it over time. I know that this can destroy a motherboard over several years of a computer not being turned on, but I don't know what the effects are for 8-16 hours of down time at a time.

i was thinking more along the lines of memory issues, with all the background programs running... back when i left my computer running my video card would trip out it seems. and when i had low memory too it would do the same.. thought it was the same way with servers

That would only be a problem if you have a program leaking memory. And even if you do, just restart that program instead of the entire system.
